  9. MATERIALS

Equipment and materials:

· Garlic root

· 1 M hydrochloric acid

· Acetic alcohol (ethanoic acid)

· Orcein ethanoic stain (acetic orcein)

· Ice cold distilled water

· Water bath at 60 ͦC

· 2 watch glasses

· Test tube

· 2 pipettes (and pipette filters)

· Microscope slides and coverslips

· Microscope

· Pair of fine forceps

· Filter paper and soft tissues

· Mounted needle (manceration)

Heath and safety:

1 M hydrochloric acid Irritant Wear lab coat, gloves and safety goggles

Orcein ethanoic Corrosive, irritant Work under fume cupboard. Wear lab coat, gloves and safety goggles

Method:

1. 2 of 1 M hydrochloric acid which was in a test tube was placed in water bath

2. 1 cm of root tips were cut from garlic root. 5 root tips were cut. They were white and firm and had rounded end.

3. Root tips were placed in a watch glass which contained 2 of acetic alcohol. Root tips were placed there for 10 min

4. Root tips were removed from the watch glass to the second watch glass which contained 5 of ice-cold distilled water. Root tips were placed there for 5 min. Then, root tips were dried on filter paper. It was important, because further precipitation may form.

5. The first watch glass from step 3 was cleaned and dried. Next, heated hydrochloric acid was put in watch glass, then, root tips were placed there for 5 min.

6. The second watch glass from the step 4 was cleaned and dried. Step 3 was repeated.

7. One root tip was transferred to the microscope slide. Then, 2-3mm from the rounded growing tip was cut. It was important to save rounded growing root tip for further steps.

8. Rounded growing root tip cells were broken up by the usage of mounted needle.

9. 2-3 drops of acetic orcein was added to the microscope slide. Acetic orcein stained for 2 min.

10. Microscope slide with root tips and cell was covered by coverslip and then bolted with several layers of filter paper. Next, coverslip was gently tapped with the end of pencil.

11. Microscope slide was placed in microscope under X400 magnification.
